Abstract This work was carried out at the Plant Tissue Culture Laboratory in the Horticulture Department, Faculty of Agriculture Suez Canal University during the years of 1997-2000 . Plants used in this work were 2 genera of ornamental trees and 2 genera of ornamental palm~ as following. 1- Bombax malabaricum De. (Silk cotton tree) Fam: Bombacaceae 2- Callistemon lanceolatus De. (Callistemon) Fam: Myrtaceae 3- Sabal palmetto Lode. (Sabal) Fam : Palmaceae (Arecaceae) 4- Washingtoniafilifera Wendl. (Washingtonia) Fam Palmaceae (Arecaceae) This work aimed to : 1- Study the effect of growth regulators as cytokinins (mainly BA) and auxins (mainly NAA) on the in vitro multiplication as well as in vitro rooting of Bombax and Callistemon to obtain the optimum regeneration phase and to develop the propagation method of these plants. 2- Study the in vitro seed germination of Sabal and Washingtonia palms, as well as study the effect of BA and NAA on the callus production on germinated seeds and shoot formation on seedling of Sabal and Washingtonia. This work included three studies: First study: Micropropagation of Bombax malabaricum . This study induced two experiments: 1- Effect of BA at (0.0, 1.0 , 2.0 and 3.0 mg/L) combined with NAA at (0.0, 0.1, 0.2 and 0.5 mg/L) on the shoot multiplication of shoot tips and node segments excised from seedlings of Bombax. 2- Effect of IBA and NAA at (0.0 , 0.1,0.2 and 0.4 mg/L) on the in vitro rooting of proliferated shoots of Bombax. The most important results were l-Application of BA at 2.0 mg/L to MS medium increased number of the proliferated shoots when both of shoot tips and node segments were used as explants. 2- Treatment with BA at 1.0 mg/L produced the tallest shoots as 4.0 cm when the shoot tips were placed into MS medium and 3.4 cm when the node segments were used as explants. 3- The root formation was induced by culturing the proliferated shoots of Bombax on MS medium with or without auxins (IBA and NAA). 4- IBA and NAA at low concentration of 0.1 mg/L increased the number of developed roots and the root length. However, the treatment with IBA gave better results than NAA. 5- The plantlets were transferred from the rooting media and cultured in plastic pots (6.0 cm) containing peatmoss : sand: vermiculite (1: 1: 1 by volume) under greenhouse conditions. The survival percentage of Bombax plantlets reached 85%. Second study: micropropagation of Callistemon lanceolatus This study induced two experiments: 1- Effect of MS salt strength (Y2 MS and full MS), BA at (0.5, 1.0,2.0 and 3.0 mglL) and NAA at (0.1,0.2 and 0.3 mg/L) on the in vitro shoot multiplication of shoot tips taken from Callistemon seedlings. 2- Effect of IBA and NAA at (0.5, 1.0 and 3.0 mg/L) on the in vitro rooting. |
