Abstract

From the results of the current study it could be concluded that all rats showed statistically significant elevations of CPK, LDH and liver function tests in both adult and senile rats after acute single oral dose administration of PDE-5 inhibitors.
Meanwhile, no significant changes were noticed in all the biochemical parameters on daily administration of the same drugs neither for 90 days nor after 30 days recovery period.
Extensive testicular atrophy and focal degeneration of the testis are frequent histopathological findings associated with tadalafil chronic administration to rats while, photoreceptor disintegration is most evident with sildenafil and vardenafil.
All the three PDE-5 inhibitors significantly improved patient scores for question 3 and 4, as well as improved IIEF domain scores compared with placebo with nearly the same efficacy.
The most commonly reported treatment-emergent adverse events with sildenafil, vardenafil or tadalafil were headache, dyspepsia, facial flushing and backache.
