Abstract

This study was designed to evaluate the possible role of Nigella sativa (NS) oil , as immuno-stimulatory and anti-cancer agent against DMBA- induced hamster buccal pouch (HBP) carcinogenesis.

The study was carried-out on eighty-five male golden Syrian hamsters divided into four main groups. In animals of DMBA-treated groups, the left buccal pouches were painted with 0.5% DMBA , 3 times/week. Animals of NS-treated groups , were given 5 mgs /day of NS oil orally. Other groups were given both NS and DMBA at the same time , 3 times/week. Group (I) : Included fifteen animals served as untreated control. Group (II) : Included twenty-five animals , were given NS only , daily for 2 weeks , then five were sacrificed , ten animals were given both NS+DMBA for 6 weeks , and the remaining ten were painted with DMBA for 6 weeks. Group (III) : Included twenty-five animals , were given both NS+DMBA for 6 weeks , then five were sacrificed , ten animals were given NS for 6 weeks , and the remaining ten were painted with DMBA for 6 weeks. Group (IV) : Included twenty animals , were painted with DMBA only for 6 weeks , then ten were sacrificed , and the remaining ten were given NS for 6 weeks. Before animals sacrificing , blood samples were withdrawn from the orbital sinus. Both buccal pouches were surgically excised , fixed in 10% neutral buffered formaline and processed for H&E and p53 immunohistochemical stains.

- Results indicated that:

1- Topical application of DMBA in HBP, induced an immunosuppression
through reduction of lymphocytes production , produced different grades
of epithelial dysplasia , and over-expression of mutant p53.It also resulted
in evident morbidity and mortality among experimental groups.

2- Administration of NS oil , significantly enhanced the immune system
through increased lymphocytes production , inhibited the development
of advanced dysplastic changes , and decreased expression of mutant
p53 , as well as a good health performance with no deaths.
