Abstract

This study was done on 40 adult albino rats; the rats were used to investigate the protective effect of melatonin on the ulcerogenic effect induced by diclofenac sodium on the stomach. The rats were grouped as 10 in each group as follows:
Group 1 (Control group), the rats had been received oral 1 ml saline daily for 2 weeks and serve as control group.
Group 2(diclofenac group), the rats had been received diclofenac sodium orally in a dose of 10 mg/kg body weight daily for 2 weeks.
Group 3(diclofenac and melatonin group), the rats had been received melatonin in a dose of 50 mg/kg body weight daily at 8am and diclofenac sodium in a dose of 10 mg/kg body weight at 2 pm daily for 2 weeks.
Group 4 (melatonin group), the rats had been received intraperitoneal melatonin in a dose of 50 mg/kg body weight daily for 2 weeks.
The stomach of the rats was opened for detection of ulcer index (UI) and histological examination, the stomach tissue samples stained with Hematoxin and Eosin, Masson’s trichrome and Van Giesson stains and then examined under light microscope. Three rats from the diclofenac group were given India ink by intra-cardiac infusion and gastric samples were processed and examined under light microscope after staining with Van Giesson stain.
