Abstract
This thesis was designed to through some light on decomposition of cholesterol by microorganisms in liquid media, which includes the following steps: enumeration and isolation of some cholesterol decomposing bacteria from different food samples. Screening of isolates for their cholesterol decomposition using well agar diffusion method and further screening of certain isolates for cholesterol decomposition in their liquid medium. Studies some factors affecting the growth and decomposition of cholesterol by selected isolates. Assaying the production of cholesterol oxidase (COD) enzyme by the selected isolates under their optimum conditions. Selection and identification of the highly active COD producing isolate. Production, purification and characterization of the purified COD enzyme. Application on the purified COD enzyme in clinical purposes.
