Abstract

concerning the scope of future researches designed to ascertain the safety margins of griseofulvin as a clinically useful anti-fungal drug free from toxic hazards:
- Future studies aiming at the precise assessment of the therapeutic usefulness of griseofulvin should be directed in the first place to verification of the scanty reports claiming fairly the anti-fungal potency of this agent by controlled wide scale experimental trials. Extensive pharmacokinetics studies embracing tissue distribution, predilictive organ uptake and localization, metabolic degradation and pathways of excretion are also essential requirements for proper adjustement of safe dosage and appraisal of its freedom from potentially toxic hazards in cases demanding repeated therapeutic courses.
On the light of the results of the present study, we recommended the following guidelines:
1- Patients on prolonged therapy with griseofulvin should be under periodic monitoring of complete blood picture, since this drug leads to anaemia.
2- Griseofulvin should not be given to patients suffering from hepatic diseases as this drug is hepatotoxic, therefore carefull history taking should be done before starting treatment with griseofulvin.
The aim of this work was to evaluate genotoxic, hepatotoxic and haematotoxic effects of griseofulvin in adult albino rats.
