Abstract

To evaluate the roles of the some apoptotic markers in HCV-positive blood donors the present study invertigated the expression of FAS and BCL_2 as an apoptotic markers on lymphocytes using flow cytometry and correlate these markers with liver enzymes and alpha fetoptein
This study was carried out on 21 blood donorsattending the blood bank of minia university hospital during the period of the april 2003 till April2004 they were classified into :group (1)comprised 40 HCV-antibodies negative blood donors ,group (II)comprised 70 blood donors with HCV-antibodies positive without liver cirrhosis ,and group (III)comprised 11 blood donors with HCV- antibodies positive with liver cirrhosis
-routine invertigations :
1.complete blood count
2. complete liver function tests
-special invertigations:
1.aloha fetoprotein assay by ELISA method
2.detection of fas (cd95)antigen on lymphocytes by flow cytometry
3. detection of intracellular BCL-2 oncoprotein by flow cytometry
The aim of this work is to study some apoptotic markers such as FAS (CD95) and BCL-2 expression on lymphocytes in hepatitis c blood donors and alpha fetoprotein as a hepatic tumour marker.
from this study it is concluded that:
Apoptosis is involved in chronic hepatic c patients. Apoptosis is increased in these patients and plats an important role in the pathogenesis of the disease.
FAS expression on lymphocytes might serve as a clinical and non invasive marker for estimating the degree of liver injury over both ALT (as it was not correlated in severe cases with liver cirrhosis) and AST (not specific for liver).
The anti-apoptotic on co protein BCL-2 is a good marker for the development of heaptcellar carcinoma in HCV- patients especially with liver cirrhosis.
