Abstract

Rotaviruses are a major cause of acute viral gastroenteritis in
infants and children but can cause gastroenteritis in all age groups.
In the present study, 168 sewage samples were collected during two
years (2006-2008) from two wastewater treatment plants (Zenin and
EI Gabal El Asfar). Wastewater samples were collected from inlet,
after primary sedimentation, after secondary sedimentation and
chlorinated effluent of the examined WWTPs. Samples were
concentrated by filtration through nitrocellulose membrane (0.45~m
pore size and 142 mm diameter) and re-concentrated by organic
flocculation method for detection of rotavirus. Also 567 stool
samples were collected from hospitalized children in Said Galal
hospital, Bolak hospital, Abo El Rish hospital during one year
period of this study. Examination of stool and wastewater samples
were carried out by using RT-PCR to detect rotavirus geno~es then
confirmed by sequencing. G and P typing was carried out by
multiplex nested PCR and multiplex semi-nested PCR.
The result showed that the incidence of rotavirus were 20.8%
and 28.6% in raw wastewater and stool samples, respectively. The
genotypes of rotavirus in stool samples were G I P[ 4j, G3P[ 4],
GIP[6] and G3P[6] in percentages of 14.8%, 9.9%, 9.9% and 8.60/0,
respectively. On the other hand rotavirus genotypes in wastewater
samples were G IP[ 4] and G3P[ 4], in percentages of 20.6 % and
5.9%, respectively.
