Abstract In this thesis, simple, rapid and sensitive analytical methods were developed for the determination of sex phenolic group containing drugs as namely terbutaline sulfate, fenoterol hydro bromide, isoxsuprine hydrochloride, etilefrine hydrochloride, ethamsylate and doxycycline hyclate. These methods aimed to the analysis of the investigated drugs either in their pure forms or in their pharmaceutical dosage forms. Part I: General introduction Includes introduction about the investigated drugs, information about their chemical structure, pharmacological ~ffects, mechanism of action and their chemical and physical properties. This part also comprises a literature review of the reported analytical metho1ds used for the determination of the I studied drugs. ’ Part II: Kinetic spectrophotometric determination of the investigated drugs via their reaction with Folin-Ciocalteau. Comprises a kinetic spectrophotometric determination of the studied drugs using Folin-Ciocalteau reagent in presence of 20 % w/v sodium carbonate. This part includes two sections: •Section A: Includes the experimental part containing information about the instruments, drugs, reagents, solutions, pharmaceutical formulations and procedures used in this method. •Section B: Includes the results obtained from the analysis of the investigated drugs by the proposed method. In addition to full details about the optimization parameters, kinetics of the reaction (initial rate •Section B: Includes the results obtained from the analysis of the investigated drugs by the proposed methods. In addition to full details about the optimization parameters, suggested reaction mechanism and the stochiometry of the reaction using the limiting logarithmic method. The results are validated statistically and through recovery studies. The methods have been successfully applied to the determination of these drugs in their commercial dosage forms. Statistical comparison of the results with reference method by means of F- and t- tests at 95 % confidence limit shows excellent agreement and indicates no significant difference in accuracy and precision. I i Simultaneous determinatilm of etilefrine hydrochloride and chlorpheniramine maleate using spectrophotometric methods. Some of the studied drugs are used in the Egyptian market m combination dosage forms, so this part contains three methods for the simultaneous determination of etilefrine hydrochloride (as an example of the phenolic drugs) and chlorpheniramine maleate. These methods are: 1.First derivative method for simultaneous determination of etilefrine hydrochloride (at 246.5 and 302.5 nm) and chlorpheniramine maleate (at 264.5 nm). 2.First derivative ratio method for simultaneous determination of etilefrine hydrochloride (at 284 and 308 nm) and chlorpheniramine maleate (at 260 and 272 nm). 3.Isosbestic point method for simultaneous determination of etilefrine hydrochloride (at 292 nm) and chlorpheniramine maleate (at 231, 253 and 273 nm). |
