Abstract

In this work some bacteriological and immunological studies on Nigella sativa (N. sativa) were carried out as for evaluation of the N. sativa antibacterial and immunomodulatory effects on the immune cells, specifically monocytes and macrophages. Blood samples were collected from apparently healthy sheep for separation of monocytes and deriving to macrophages.
At first, antibacterial effect of black seed oil was tested against Staphylococcus aureus to confirm the antibacterial action and then in vitro effect on Monocytes-derived Macrophages (MDM) inflammatory functions.
So, at the beginning culturing of Monocytes-derived Macrophages (MDM) and this method has described a simple reproducible method for obtaining a high yield viable monocytes and developing them into functional macrophages in a short period of time (about 5 days). The MDM have the typical morphologic and functional characteristics of tissue macrophages and kill intracellular organisms as confirmed in these studies in bovine models.
