Abstract

The present work dealt with studying the phototoxic effect of different concentration of hematoporphorin dihydrochloride (HPD) in the presence of laser on several rheological factors and biophysical parameters of normal human blood.
To evaluate the role of different types of laser on the blood, blood viscosity, dielectric properties, hemolysis and viability test were investigated. Finally laser mechanisms which cause these effects were discussed.
Different concentration of hematoporphorin dihydrochloride (HPD) were prepared (from 0 -100 µM) and incubated with blood samples for two hours at 37 oC in dark.
Then blood samples were divided into four main groups:
Twenty samples were irradiated with infrared laser for 1 min.
Twenty samples were irradiated with YAG laser for 1 min.
Twenty samples were irradiated with visible diode laser for 1 min.
Twenty samples were not irradiated with laser and used as control group.
Each main group was divided into two subgroups:
Subgroup I used for measuring hematological parameters, dielectric properties and viscosity.
Subgroup II erythrocytes (red blood cells) obtained from the whole blood was used for analysis of hemolysis and viability test.
II- from this study we can conclude that:
A decrease in Hg concentration (g/dl), Hematocrit percent (Ht %) and Red blood cells count (RBC’s count) appeared with the increasing of photosensitizer concentration with and without laser irradiation at 904, 532 and 650nm.
The increases of concentration of hematoporphorin dihydrochloride (HPD) increasing the relative viscosity with and without exposure to laser.
Increases in photosensitizer concentration causes increasing in blood mortality with and without exposure to laser.
Increases in hemolysis of erythrocytes (RBC’s) with increasing of HPD concentrations appeared.
Irradiation of the blood suspension with infrared, YAG and visible laser in the presence of photosensitizer decreases the real values of permittivity and increased the values of conductivity at all frequencies.
Electrical impedance decreased by the increase of photosensitizer concentrations.
Increasing of the photosensitizer concentrations causes decreases in the real permittivity increment and the maximum imaginary permittivity values.
Change of real permittivity and conductivity could be due to the rupture of erythrocytes (RBC’s) membrane, exiting of the cell content to suspended medium and cell death.
