Abstract The present study aimed to evaluate three types of Vero cell rabies vaccines; French, Chinese and the experimental Egyptian one. The aspects of this comparative evaluation are safety, sterility, purity, potency, stability and the immunogenicity of each one. The test for residual live virus using mice inoculation assay showing that the three test .vaccines were completely free from any residual live virus. Test vaccines were sterility test confirmed and no microbial growth was detected on culture media. Vero cell antigen content was evaluated in test vaccines using ELISA. It was the least in the French product followed by the Chinese and the Egyptian respectively. Also the DNA content was of minimum value in the French and Chinese vaccine followed the Egyptian one. Potency of test vaccines was evaluated using NIH assay. The French vaccine showed the highest potency value followed by the Chinese and the Egyptian vaccines respectively. All of them were accepted as potent vaccines according to the WHO regulations. Thermal stability of test vaccines was evaluated post exposure to different temperatures and the relative potency revealed a good stability of test vaccines at 37oC, but a depletion of the relative potency values of the Chinese and Egyptian vaccines post exposure to 45°C was revealed, while the French vaccine potency still within the accepted limits. Immunogenicity of the French vaccine produced a much better immune response followed by Chinese and Egyptian experimental one respectively. Also, results showed that the I1m rout of vaccination is more immunogenic than the Sic rout. |
