Abstract

investigation was to investigate the S63 Inbred line (Male inbred line), S7 Inbred line (Female inbred line), S.C.10 controlled produce with researcher, S.C.10 produce with researcher according the traditional method for hybrid seed production and S.C.10 commercial seeds. Using morphological and biochemical methods of assessment to furnish standard uniform and definitive information of the verification of varietal purity in maize seed using identification characteristics of the genotypes.
This study was carried out during 2009 and 2010 seasons at the Farm of Faculty of Agriculture, Mansoura Univ., and Laboratories of Seed Tech. Res. Sec., Field Crops Research Institute, ARC. The aim of this study was to estimate the varietals purity in SC10 from three different sources.
A randomized complete blocks design with four replicates were used.
I- Field experiments: The morphological identification was conducted according to (UPOV).1-Qualitative characters 2- Quanitative characters.
II- Laboratory experiments:
1- Germination characters:first count,Germination percentage ( % ),Germination rate,Shoot length (cm)and Root length (cm).
2- Randomly Amplified Polymorphic DNA (RAPD-PCR).
G3 was the belongest one to the society in attitude of blade on leaf just above upper ear, anthocyanin coloration of brace roots, anthocyanin coloration of glumes, anthocyanin coloration of fresh anthers, spikelets density, length of peduncle and shape of ear. It gave insignificant differences in all characters except in shape of ear and shape of tip of the frist leaf it gave significant differences
G4 was the belongest one to the society in shape of tip of the first leaf and shape of ear. It gave insignificant differences in all characters except in anthocyanin coloration at base of glum it gave significant differences G5 was the belongest one to the society in anthocyanin coloration of sheath, anthocyanin coloration at base of glum, intensity of anthocyanin coloration of silks, attitude of lateral branches and anthocyanin coloration of sheath in middle of plant. It gave insignificant differences in all characters except in shape of ear it gave significant differences
The t.test in pairs was observed that the differences between every two genotypes (G3-G4; G3-G5 and G4-G5) were not significant in all characters. The dendrogram based on RAPD analysis separated the studied genotypes into two main clusters, the first cluster included all the studied genotypes except G1 was in a separate cluster.RAPD can identified markers for cultivars 4 and 5. Cultivar 5 has two cultivar-specific RAPD markers at 904 and 303 b.p. of primers A03 and B11 while, cultivar 4 has one marker at 1327 b.p. of primer A03.
from this study It could be summarized that seed has been produced under a seed certification program are the best for high grain yield due their varietals purity and genetic purity. The verification of genetic purity, seed quality characters and the identification of studied maize genotypes under the environmental conditions of Dakahlia Governorate.
