Abstract The disposition kinetics of florfenicol in either alone or combined with flunixin meglumine were studied following a single intravenous and intramuscular injections in dairy goats. The effect of drug given alone or combined with its therapeutic potentiator on some serum enzyme activities and on some blood constituents were studied following i.m injection for five consecutive days. The concentrations of drug in serum and different body fluids were determined using HPLC. Flunixin meglumine alters pharmacokinetics of florfenicol after i.v and i.m injections and affect its rate of excretion in urine, milk and ruminal juice. Florfenicol-flunixin combination caused a significant increase in alkaline phosphatase and ALT activity and a significant decrease in creatinine and urea levels, while florfenicol only caused a significant increase in AST, alkaline phosphatase, creatinine and urea levels. The ratio between florfenicol clearance from blood of goats to creatinine clearance following i.v and i.m injection of florfenicol alone and florfenicol-flunixin combination was less than one indicating that the glomerular filtration is the main pathway of eliminating florfenicol through kidneys. The data obtained showed that after i.v injection of florfenicol alone or its combination with flunixin meglumine there was a reduction in ruminal gas fermentation at 1 and 2 hours post- injection. While there were no effect after injection of florfenicol and its combination intramusculary. |
