Abstract

The aim of the present study is to clarify the effect of topiramate and fish oil administration on monoamines and amino acids in brain, in addition to amino acid in serum, cerebrum functions as well as oxidative stress and antioxidant defense system in brain, liver and testis, sodium, potassium, LH, FSH, testosterone concentration in serum of epileptic rats.
The obtained data were subjected for statistical analysis and revealed the following:
1) The treatment of epileptic rats with topiramate or fish oil produced decrease in the monoamines levels.
2) The treatment of epileptic rats with topiramate or fish oil induced a significant decrease in the aspartate and glutamate concentration in the brain and a significant increase in the glycine and GABA concentration in the brain.
3) The treatment of epileptic rats with topiramate or fish oil produced a significant decrease in the glutamate and aspartate concentration in the serum and a non significant increase in the glycine and GABA and concentration in the serum.
4) With regards to the oxidative stress and antioxidant defense system, lipid peroxidation and endogenous antioxidative enzymes in brain, liver and testis were profoundly elevated in the epileptic rats. However, the changes of glutathione contents in brain, liver and testis of these animals were decreased. The treatment with topiramate or fish oil produced an increase in the glutathione contents in the brain, liver and testis and a significant decrease in the lipid peroxidation products, all antioxidant enzymes in the brain, liver and testis
5) The treatment of epileptic rats with topiramate or fish oil produced increase in sodium concentration in serum and decrease in potassium concentration in serum.
6) With regards to sex steroid hormones, they were commonly decreased in epileptic rats. The treatment of these animals with topiramate or fish oil induced a significant increase of the diminished values of these parameters.
7) The treatment of epileptic rats with topiramate or fish oil produced decrease in total creatine kinase, AST and lactate dehydrogenase activities in the serum epileptic rats, but, ALT activities were not significantly affected in all tested groups.
In conclusion, the present study provides evidence that the topiramate and fish oil induced a potential increase of the lowered LH, FSH and testosterone levels. Also, the present study high lights that the amelioration in the altered hippocampal neurotransmitters, brain oxidative stress and antioxidant defense system as a result of treatment of epileptic rats with topiramate or fish oil may have an important role in neuroprotection against pilocarpine induced epileptic model. In order to identify the mechanism(s) of action of fish oil components further chemical and molecular research is needed to identify which of the many oil components is responsible for the observed effects. However, further clinical studies are required to assess the safety and efficacy of this oil in human beings.
