Abstract

The object of this study was to prepare and improve some inactivated RHDV vaccines using recently isolated local field strain which identified through HA test, HI test (alpha procedure), RT/ PCR and the PCR product was sequenced and submitted to GenBank under JX312065 as accession number and RHDV2010 as name. Alignment of RHDV2010 and Giza 2006 revealed 98% nucleotide identity. The RHDV2010 used in the preparation of inactivated lyophilized RHDV vaccine reconstituted in 6 cm saline, lyophilized vaccine reconstituted in 6 cm DEAE- Dextran substance as immunostimulant, Oil RHDV vaccine and Oil vaccine with DEAE-Dextran as immunostimulant. In order to evaluate the immune response and duration of immunity the prepared vaccines inoculated into 4 different groups the inoculated rabbits observed for six months. When the obtained HI- antibody titre in the vaccinated groups was compared it revealed that group I had the lowest HI titre as the peak was 6.2 Log2 at 21st DPV and decreased gradually, with 80% protection. While the obtained HI- Antibody titre in the other 3 vaccinated groups were related with 100% protection and the HI- antibody titre reach the highest 11.2 Log2 at 180th, 10.8 Log2 at 120th and 11.2 Log2 at 120th DPV with the group II, III &IV respectively. While dealing with cellular immune response the optical density (ΔOD) of lymphocyte blastogenisis recorded the highest at 27th DPV as 0.988, 1.869 and 1.621 with group I, III&IV respectively while group II recorded 1.084 at 13th DPV. While the phagocytic activity record maximum phagocytic % at 6th DPV 62.5, 77, 85 and 85% with group I, II, III& IV respectively. While phagocytic index recorded 0.8, 0.71 and 0.83 with group I, II&III and group IV gave the highest index in compare with other groups as recorded 1.0 at 13th DPV.
