Abstract The present study attempted to investigate the possible protective effect from gastric ulceration of some antioxidants like melatonin and hydrogen sulphide and the possible potentiation of the melatonin effect by co-administration with pyridoxine. The present study used cold restrained stress model as a well-established model of gastric ulcer induction. Male albino rats were fasted for 48 hours with free access to water prior to the experiment except for the last hour, rats deprived from water. Rats were pylorically ligated under light ether anesthesia. After pyloric ligation, animals were restrained by fixing the four limbs to a wooden board and they were placed in refrigerator at 4 ± 1 OC for 3.5 hour.The study involved the following experimental groups:1- Control non-stressed group; rats were left freely wandering in their cages for 3.5 hours after being subjected to pyloric ligation. 2- Cold restraint stress group (CRS); rats received 0.05% tween80 only i.p forty five minutes before CRS. 3- CRS + Melatonin group; rats received 15 mg/kg of melatonin in 0.05 tween80 i.p forty five minutes before CRS. 4- CRS + Melatonin + pyridoxine group; rats received 15 mg/kg melatonin in 0.05% tween80 i.p and 10 mg/kg pyridoxine i.p forty five minutes before CRS.5- CRS + pyridoxine group; rats received 10 mg/kg pyridoxine i.p forty five minutes before CRS. 6- CRS + NaHS group; rats received 60 micromol/kg Sodium Hydrogen Sulphide i.p forty five minutes before CRS. |
