Abstract

Enumeration and isolation of cholesterol growing bacteria from yoghurt product were attained and identified and confirmed identified were based on alignment of 16S rRNA gene sequence in gene bank. Sixty five of the bacterial isolates were screened for cholesterol decomposition. The degree of acid and bile tolerance of bacteria was evaluated. Lactococcus lactis KF 147 was able to maintain viability for 2 h at pH 2 and to grow in a medium with 4,000 mg of bile acids per liter with maximum cholesterol reduction 66.8 and 58.4 % for L. lactis KF 147 and E. faecalis DAPTO 512 in liquid medium, respectively. Then the bacterial isolates screened for bile salt hydrolase and cholesterol binding activity. The optimal factors for the bacterial growth and cholesterol reduction were studied. The growth of bacterial isolates affected by the presence of cholesterol in medium. The growing cells of the bacterial isolates were cholesterol reducing activity more than resting and dead cells. Transmission electron microscope was studied to compare between shapes and size the cells of bacterial isolates with and without cholesterol. IR analysis was detected the functional groups attached binding between cholesterol particles and surface of the cells it indicated that, the cholesterol binding activity between the isolates and cholesterol. The fatty acid composition of the cells of L.lactis KF 147 was altered by reducing cholesterol from the media that indicated incorporation the cholesterol particles into cell membrane of cells. According to these results, cholesterol reducing bacteria has revealed the probiotic potential in hypocholesterolemic effect on rat with hypercholesterolemia induced by diet were affecting on body weight, feed efficiency, plasma lipid profile and feces samples of rat.
