Abstract

This work was carried out to study the occurrence of Cryptosporidium in feces of calves, lambs and goat kids in veterinary clinics, farms and house holds animals. A total of 546 animal faecal samples and 160 human stool samples were collected from rural settings in Cairo, Giza and Beni-Sweif governorates. The incidence of Cryptosporidium infection was 73 (41.7%) in cattle calves, 41 (33.3%) in buffalo calves, 47 (36.7%) in lambs, 48 (40%) in goat kids and 0% in human using modified Ziehl-Neelsen staining.Meanwhile,ELISA results for some of examined samples evidenced that 17 of 31 (53%) in cattle calves, 20 of 31 (64.5%) in buffalo calves and 2 of 10 (20%) in lambs and goat kids and 8 of 91 (8.8%) in human samples were positive. Application of multiplex PCR on 14 positive fecal bovine samples found that 7samples were mixed infection with Cryptosporidium spp. and 7samples have single infection; 5 of them were C.parvum and the other two were C.andersoni. Moreover study on the viability of Cryptosporidium oocysts found that heating at 100ºc for 1min or more, freezing at -20ºcfor 24h, chlorine with concentration 8ppm and filtration in filters of pore size less than 1µm have effect on oocysts viability.Keywords:Cryptosporidium-calves-lambs-goat kids-human-viability-ELISA- multiplex PCR.
