Abstract

The present study was performed on 25 testes of adult
buffalo bull. For light microscopy the samples were fixed in 10%
buffered neutral formalin and processed until sections of 5-7!lm in
thickness were obtained and stained with different stains. For
electron microscopy, the specimens were fixed in 3%
glutaraldehyde solution in O.l M sodium phosphate buffer pH 7.4 at
4°c for 3 hours and post fixed in 1% osmic acid. Sections were
stained with uranyl acetate and lead citrate, examined and
photographed with with JEOL JEM-IOlO TEM operating at 80 kv.

The testicular weight of adult buffalo bull ranged from 225-
250 gm, its width was of about 4-5 cm and its length of about 7-8.5
cm. The testis of the buffalo bull was lain in scrotum with a vertical
longitudinal axis, the head of epididymis was situated dorsally,
while the tail was ventrally, The epididymal border of the testis was
located caudally and the free border was cranially
The histological finrlings:-
Each testis was enclosed by white fibrous capsule; tunica
albuginea, composed mainly of collagen fibers, the connective
tissues extend into the organ, forming a mass, called the
mediastinum testis. The seminiferous tubules were divided into
tubuli contorti (convoluted seminiferous tubules) and tubuli recti
(straight seminiferous tubules) which connect the tubuli contorti to
the rete testis. The lamina propria of seminiferous tubules was
limited by PAS positive basal lamina. Under EM the basal lamina
