Abstract This work was carried out to investigate the microbiological and parasitological relationship (E.(;oll, Salmonella and Eimeria) in healthy and diseased birds in commercial broiler chickens and some layer flocks in Kafr El-Seikh and Behera governorates. The pathogenicity of E. coli isolates, effect of chemotherapeutic agents on E. coli isolates and characterization of isolated Salmonellae were studied. Parasitic infestation was done by microscopic examination of intestinal scrapings. The results and conclusions are summarized in the following: 1- Out of 24 broiler flocks and 4 layer flocks: 14 E. coli isolates and 6 Salmonella isolates were recovered while Eimeria sp. were present in 10 flocks from 24 broiler flocks. 2- The 14 E.coli isolates were Congo Red E.coli positive on Congo red media and revealed 92.85, 91.83 % in embryo infected with 10 3 and 104 CFU respectively. 148 ;’..” Summary 3- In-vitro sensitivity test of E. coli isolates revealed that ciprofloxacin, norfloxacin possessed the highest activity against E.coli (92.85%), then gentamycin and neomycin (64.28%), chloramphinicol moderately affect (57.14%) and colistine (35.7). Also all 14 tested isolates except one showed resistance patterns to streptomycin, amoxycillin, and doxycycline while all isolates were resistant to penicill in. 4- The six Salmonella isolates were serotyped as follows: 2 isolates S.enteritidis (33.33%); 3 isolates S.typhimurium (50%); and one isolate untyped Salmonella (16.66%). 5- In-vitro sensitivity test of Salmonella isolates revealed that all isolates were highly sensitive to ciprofloxacin, norfloxacin and streptomycin (83.3%); amoxycillin (66.66%); and moderately sensitive to neomycin (50%); gentamycin and chloramphenicol (33.33%); and colistine (16.6%). Meanwhile, all isolates were resistant to doxycycline and penicillin (100%) 6- Detection of plasmid revealed the appearance of plasmids in 6 Salmonella isolates with similar molecular weights (6Kbp). 149 Summary 7- Sodium dodcyl sulphate - polyacrylamide gel electrophoresis (SDS-P AGE) of different salmonella serotypes showed that there are differences in the protein make up of different serotypes. 8- Intestinal scraping of 4 parts of intestine revealed that all Eimeria sp. detected were cecal coccidiosis only. 9- There are some correlations between parasite detected (Eimeria sp.) and bacterial isolates (E. coli and Salmonella) especially in broiler flocks as follow: • E.coli and Eimeria species correlations were 25% In the group I (14-21 days old) and 37.5% in the group II (22-30 days old) . • Salmonella and Eimeria species correlations were 12.5% In both group II (22-30 days old) group III (32-42 days old). • E. coli and Salmonella species correlations were 12.5% in each of group I (14-21 days old) and group II (22-30 days old). |
