Abstract

Different types of bones from different animals had been collected to test the effect of different fixatives and decalcifying agents on the bone tissue and on its affinity to different stains. Strong acids like hydrochloric acid (Jenkin,s fluid) and nitric acid (Perenyi,s fluid) resulted in rapid decalcification but showed swelling of collagen of bony lamellae and impairment of staining. 5%Trichloroacetic acid showed rapid decalcification of bone with minimal harmful effect on tissue with good staining affinity of bone sections. Formic acid and neutral E.D.T.A. exhibited slow decalcification process with preservation of tissue structures that showed high affinity to different stains. Mϋller,s fluid enhanced the process of decalcification.10% neutral buffered formalin and Bouin,s fluid are satisfactory fixatives for bone fixation before decalcification process. Helly,s fluid delayed the decalcification process. The raised temperature (40oC.) accelerated the process of decalcification but caused maceration and fragmentation of bone fibers and resulted in impairment of staining.
