Abstract

Stem cells have the unique ability to divide and to make copies of
themselves (self-renewal) and to other more specialized cell types through
differentiation under the appropriate conditions, and most importantly having
cluster of differentiation (CD34+) in human and (SCA-l +) in mice. Stem cells that
form blood and immune cells known by hematopoietic stem cells (HSCs).
Hematopoietic stem cells transplant are now under investigation for using to treat
patients with chronic and incurable diseases such as Parkinsonism and Alzheimer.
Differentiation of HSCs into immune cells could help in controlling of
immunodeficiency and in vaccine or drug delivery and in solving medical
problems related to immune system. Differentiation of HSCs to immune cells still
limited and no sufficient studies had been done in this field.
In this study, efforts were exerted in a trial to differentiate HSCs into
immune cells. Samples were collected from human umbilical cord blood in
anticoagulant collecting tubes directly after the female donors giving birth. The
HSCs were isolated by depending on their specific cell surface marker (CD34+),
and the following was done in sequence.
A. Isolation of CD34+ cells:
In this study, HSCs were isolated using magnetic cell sorting based on
positive selection of CD34+ cell marker, as the cells were treated with CD34+
progenitor cell isolation kit and pass through OctoMACS system several times to
confirm their purity.
