Abstract

This study was designed to evaluate the effect of olive leaves and oil on some biochemical constituents in rabbits kept on high sucrose ration.
Study lasted for three months and was carried out on a total of 40 male Newzeland rabbits of 1300- 1600gram body weight and about 50±10 days old that were divided equally into four groups ( 10 rabbits) per each group as follow:
1- First group (Group І) : served as control and kept on basal ration only.
2- Second group (Group ІІ ) : received basal ration of 30% sucrose concentration.
3- Third group (Group ІІІ) : received basal ration of 30% sucrose concentration and olive leaves (5% /kg ration).
4- Fourth Group (Group ІV ) : kept on basal ration of 30% sucrose concentration and olive oil (5% /kg ration).
All treatments were supplied for two months then they were stopped for one month.
The blood samples were collected from all animals under experiment three times, one after 30 days(from ear veins), while the second one after 60 days and the last one after 30 days from withdrawal all treatment by slaughtering the animals.
Blood samples were collected from ear veins in clean and dry Wassermann tubes . Serum was separated for determination of:
-Serum liver functions( AST, ALT,GGT and ALP).
- Serum protein pattern (total protein, albumin, globulin and A/G ratio).
-Serum lipid profile(total lipid,TAG,total cholesterol, HDL-c LDL-c, VLDL-c ).
-Atherogenic indices.
-Serum Testosterone.
-Serum zinc.
-Serum copper.
-Kidney functions (urea and creatinine).
-Blood Glucose.
Liver, Heart and kidney tissues were collected for evaluation of:
- Liver weight.
- Reduced glutathione, lipid peroxidation, hydrogen peroxide and antioxidant enzymes ( Catalase and Glutathione –S- transferase ).
Heart, kidney and liver tissues were taken for:
-Tissue histopathology.
Our results and Statistical analysis revealed the following:
1- High sucrose fed ration revealed significant increased in most serum liver enzymes activities .
2-Treatment of high sucrose fed rabbits with olive leaves or oil revealed significant decrease in liver enzymes.
3- Our results also revealed that, feeding olive oil or leaves don’t ameliorate decreasing effect on (TP , Albumin and A/G) of high sucrose diet in animals.
4- High sucrose fed ration in rabbits induced hyperlipidemia. As, it significantly increased serum total lipids, TC, TAG, LDL-c while, it significantly decreased HDL-c as compared to control group.
5- Treatment of high sucrose fed rabbits with olive leaves or oil revealed significant decrease in hyperlipidemia. Inaddition these results
revealed that, olive leaves and oil success in ameliorate the atherogenic effects of high sucrose diet.
6- Significant increase in testosterone level in group kept on high sucrose diet, but it decrease in groups treated with olive leaves or oil.
7- The obtained results revealed that, high sucrose diet decrease serum zinc concentration.
8- Treatment of high sucrose fed rabbits with olive leaves and oil revealed significant alleviation in decreasing zinc level.
9- The obtained data referred that, significant decrease in blood urea and serum creatinine level in group fed on sucrose plus olive oil in compared to other groups after 30 and 60 days of treatment .
10- The prolonged high sucrose diet showed significant increase in blood glucose level even after withdrawal time.
11- Treatment of high sucrose fed rabbits with olive leaves or oil revealed significant hypoglycemic effect only after withdrawal time.
12- The obtained results indicated significant decrease in feed consumption in all groups if compared with control group.
13- Our results showed a significant decrease in liver weight in control group and group fed on sucrose plus olive leaves in compared to sucrose fed group and sucrose plus olive oil fed group after 60 day of treatment .
14- In liver tissue of rabbits kept sucrose high diet possess the highest values of hepatic MDA and H2O2, while it possess the lowest levels of hepatic GSH, Catalase and GST . Most of these criteria were greatly improved to these in control group after adding of olive leaves or olive oil to sucrose.
15-In heart tissue, the highest values of GSH were noticed in rabbits kept on olive leaves followed by that kept on olive oil , while the lowest values was observed in those kept on sucrose . On the other hand, the highest values of MDA and H202 were recorded in sucrose fed group. While the same group
possess the lowest values of catalase and GST and the feed of olive leaves and oil success for extent in increasing the values of there two enzymes.
16- The sucrose feeding only significantly increased kidney GSH, MDA ,and H2O2,while it significantly decrease the activity of both catalase and GST level they still less than those in control group. The addition of olive leaves and oil decreasing the values of MDA and H2O2 towards those of control group.
17- Histological criteria demonstrated a marked fatty changes in liver of rabbits fed on high sucrose diet and sever hydrobic degeneration . In contrary treatment by olive leaves and oil success in alleviate liver fatty changes .
18- Moreover there were marked tubular necrosis and hemorrhage in kidney of rabbits fed on high sucrose diet. On the other hand olive oil and leaves decrease this changes.
19- In heart histology revealed that, sucrose fed rabbit showing lytic necrosis with inflammatory cells and erythrocytes infiltrated. In contrary olive leaves and oil not revealed these lesions.
