Abstract

The thesis deals with the quantitative determination of some pharmaceutically active compounds having sulphur atom in their chemical structures. These compounds belong to different pharmacological classes and therefore have different therapeutic uses. For instance mesna is used as an adjuvant therapy in cancer treatment, acetylcysteine is used as a mucolytic, timonacic acid is useful in treating hepatic disorders and sulpiride is used as antipsychotic. The thesis consists of five chapters:
Chapter I
This chapter includes a general introduction regarding chemical names, structures, molecular formulae and molecular weights, physical properties, pKa, pharmacological properties and therapeutic uses of the six investigated drugs. It also contains the commercial products or the laboratory prepared dosage forms used throughout the thesis.
Chapter II
Chapter II includes a literature review of the methods that have been reported for the analysis of mesna in bulk powder, pharmaceutical dosage form as well as in biological fluids.
This chapter describes spectrophotometric approaches for the quantitative estimation of mesna in bulk form and in its commercial ampoules. It is divided into two sections:
Section (A):
In this section two nucleophilic aromatic substitution reactions were proposed for the determination of mesna through derivatization with 4-chloro-7-nitrobenzo-2-oxa-1,3-diazole (NBD-Cl) (Method I) and 2,4 dinitrofluorobenzene (DNFB) (Method II). The reactions took place in the presence of 0.05 M borate buffer of pH 8.5 (for method I) and 9.5 (for method II) and the produced colored derivatives were measured after 30 minutes at 414 and 332 nm for method I and II, respectively.
