Abstract This work aimed to isolate and identify L.monocytogenes from different food stuffs and water. In addition, to evaluate the accuracy of polymerase chain reaction and conventional methods of diagnosis. EL-Sharkia governorate was a scope of the present study . This study was conducted on 100 different food stuffs samples and 50 fresh water samples which surveyed for presence of L. monocytogenes. The food stuffs were dairy products including raw unpasteurized milk, pasteurized milk, Kareesh cheese and meat products including luncheon, minced raw meat and beef. All tested samples included 100 different food stuffs from (milk, cheese and meat) and 50 water samples were inoculated into two enrichment media then cultivated on Oxford agar. Presumptively positive isolates were determined by colonial morphology, Gram’s stain, catalase test, haemolysis on sheep blood agar and motility test. The isoates were further identified to species level by 10300 API Listeria strips. PCR was done for all specimens to evaluate its accuracy. Antimicrobial susceptibility was done for all isolates was also done. |
