Abstract

commonest diseases nowadays in both developed and developing countries. It consists of a group of risk factors as central obesity, Insulin resistance that develops to diabetes, hypertension, dyslipidemia in the form of hypertriglyceridaemia, hypercholesterolaemia and fatty liver. It is due to shift toward a more sedentary lifestyle accompanied by disturbed energy utilization and increased caloric intake. High fructose intake, considered as a highly lipogenic factor, may be an important causative factor in the development of metabolic syndrome due to activation of NF-κB inflammatory pathway. In the present study, high fructose diet (65%) for 8 weeks was used to induce MetS in rats. Forty male albino rats, weighing 150-200 gm were used. Rats were divided into four groups, each consists of 10 rats: Group 1 (control): Rats were fed a chow diet for 8 weeks. Group 2 (vehicle treated): Rats were fed a fructose enriched diet 65% (FED for 8 weeks) and ethyl alcohol 20%. Group 3 (resveratrol treated): Rats were fed a fructose enriched diet 65% (FED for 8 weeks) and received at the same time resveratrol in a dose of 10 mg/Kg dissolved in ethyl alcohol 20%. Group 4 (quercetin treated): Rats were fed a fructose enriched diet 65% (FED for 8 weeks) and received at the same time quercetin in a dose of 50 mg/Kg dissolved in water. The following parameters were measured: Rat tail systolic blood pressure and measurement of body weight. Laboratory investigations: measurement of serum TNF-α, leptin, glucose, insulin, Calculation of HOMA-IR, triglycerides, Cholesterol, Uric acid, catalase and MDA. Histopathological examination: The liver by hematoxylline & eosin and immunohistochemistry using NF-κB. The results showed increased body weight, systolic blood pressure, serum TNF-α, serum leptin, seum glucose, serum insulin, HOMA-IR as an indicator of insulin resistance, serum triglycerides, serum cholesterol, serum MDA, serum uric acid, pathological picture of steatohepatitis by hematoxylline & eosin and increased NF-κB expression in vehicle treated group. Both resveratrol and quercetin improved these parameters but quercetin improved these parameters better than resveratrol as compared to vehicle treated group.
