Abstract

Scope of investigation
The studied antihypertensive drugs (namely; AML, FLD, LDP, NDP, MTP and RMP) , are pharmaceutically important compounds that are extensively used in Egypt and in many countries worldwide in the treatment of hypertension and other cardiac diseases.
The aim of this work was designed for:-
•Development of simple, selective and accurate TLC-densitometric method for simultaneous determination of FLD - MTP and FLD - RMP mixtures in their dosage forms.
•Development of rapid, simple and sensitive micell-enhanced spectrofluorimetric methods for the determination of AML, FLD, LDP and NDP with no need for derivatization reactions.
•Study the possible degradation product(s) of AML after exposure to different stress conditions (acid, alkali, oxidation and light) using the proposed micell-enhanced spectrofluorimetric method as a stability indicating assay.
•Application of the proposed methods for determination of the studied compounds in pure form and different pharmaceutical dosage forms.
•Determination of AML, FLD ,LDP and NDP in spiked human plasma using the developed micell-enhanced spectrofluorimetric method with no need for extraction or pretreatment steps.
•Statestical comaprison for the results that have been obtained by the proposed methods with those obtained by a reference methods.
•Validation of all the proposed methods according to ICH guidelines
Results
•A new TLC-densitometric method was developed for simultaneous determination of felodipine with either metoprolol or ramipril in fixed dose combinations.
•Rapid, Simple and highly sensitive spectrofluorimetric methods were developed for determination of FLD, LDP, AML and NDP, with no need for derivatization reactions compared with the reported spectrofluorimetric methods. By virtue of rapidity, simplicity and high sensitivity of the proposed spectrofluorimetric methods , it could be applied to the analysis of the studied drugs in their dosage forms and in spiked human plasma.
