Abstract

The thesis is divided into two Parts:-Part I: Phytochemical and biological investigation of Ceratonia siliqua L. endophyte. Chapter 1: Cultivation of Penicillium citrinum Thom and preparation of culture media extracts. Chapter 2: Investigation of culture media extracts. Chapter 3: characterization of the isolated compounds Chapter 4: Biological study of the isolated compounds. In this parts, the phytochemical investigation of the ETOAc extracts of both rice and white bean culture media of Penicillium citrinum Thom endophyte isolated from Ceratonia siliqua L. fresh stems led to the isolation and identification of 6 new and 13 known compounds. The new compounds include: 6-methylcurvulinic acid, 1,2,3,11b-tetrahydro-quinolactacide, Citriquinochroman, Tanzawaic acids G and H and 3,5-dimethyl-6-hydroxy-8-methoxyisoquinoline. Citriquinochroman showed cytotoxicity against the L5178Y mouse lymphoma cell line with an EC50 value of 6.1 μM. Part II: Phytochemical and biological investigation of the Aglaia duperreana Lour flowers. Chapter 1: Investigation of the flowers extract. Chapter 2: characterization of the isolated compounds. Chapter 3: Biological study of the isolated compounds. In this part, the phytochemical investigation of the methanolic extract of Aglaia duperreana Lour flowers led to the isolation and identification of a new rocaglamide derivative (C-1-O-acetyl-3’-hydroxy-didesmethylrocaglamide) and twelve known congeners of which C-3’-hydroxydidesmethyl-rocaglamide was isolated for the first time from A. duperreana. Lour. All the separated rocaglamide derivatives are potent cytotoxic agents against the L5178Y mouse lymphoma cells with EC50 values ranging from (5.1 to 54.8 nM) concentration. Flavan-3-ol derivatives are also isolated and identified. Catechin is reported in the present study for the first time from A. duperreana species, while epicatechine is reported for the first time from the flowers of the same plant.
