Abstract Of our study is to isolation and characterization of the lytic bacteriophages Specific to clinical P. aeroginosa and p. mirabilis isolated from diabetic foot patients and test phage efficacy as an alternative therapy For local treatment. To evaluate therapeutic potential of phages specific for P. aeroginosa and p. mirabilis to resolve wound infection in diabetic foot rabbits. Back ground: Diabetic foot ulcer is one of the most distressing complications of diabetic Mellitus. Several microorganisms are implicated in the disease causing severe damage with many exhibits a high degree of resistance to broad spectrum of antibiotic. In this study samples were collected from 50 diabetic foot ulcer Patients. 123 Cultures were biochemically identified and antibiotics susceptibility was done. The isolated multidrug resistant P. aeruginosa and p. mirabilis, strain were further tested against lytic phages designated as P. aeruginosa and p. mirabilis phage (psA1,psA2)and (prM),Lytic phages were isolated from sewage, here we describe the isolation and characterization of lytic phages capable to infect.p. aeruginosa and p. mirabilis isolated from diabetic foot patient. Excision wound model was used. Group I was assigned as control, Group II infected with pseudomonas earuginosa , Group III non infected and received only phage preparation to assess whether phage initiates infection,Group IV infected with pseudomonas earuginosa, once the infection was established it was challenged with pseudomonas earuginosa phages and Group V infected Pseudomonas earuginosa and treated with antibiotic Amikacin after 4th day of infection.and Group VI infected pseudomonas earuginosa and recived p. earuginosa phages(psA1,psA2) and Amikacin antibiotic after 4th day of infection . Results: The phages (psA1,psA2) were studied in detail due to their efficient lysis of wide wide spectrum on Pseudomonas aeruginosa, strains.phages (psA1,psA2) morphologically classified into the order Caudovirales and family Myoviridae morphtype A1 (an icosahedral head with diameter 87.5mm and long contractile sheathed tail with diameter 147.8mm and 27.6mm neck for (psA1),while for (psA2)with head 89.7mm and 137.9mm for tail.to The order Caudovirales; Myoviridae family and phage growth cycle with a detected latent period of 15 min,and brust size of 182 plaques forming units per infected cell These phages has narrow host range and shows the remarkable thermal resistance and is viable up to 60 ºC. Sincere efforts in term of identification, isolation, purification and characterization of multiple types of phages against Pseudomonas aeruginosa and proteus mirabilis. The maximum adsorption values for phages (psA1,psA2) were adsorbed after 30 minutes and for(prM) 20 minutes. The one- step curve was evaluated for the isolated phages and the values of the latent period of (psA1)and (psA2)were found to be 15 min and 20 min respectively, followed by a rise period lasted at 25 min and 30 min respectively,and the average burst size was approximately ~182 and ~ 76 pfu per infected cell respectively. A significant decrease in infection, period of epithelization and wound Contraction was observed in the phage challenged group and phage and Antibiotic group when compared to antibiotic treated diabetic rabbits and the control group. Conclusion: To conclude the study provide new insights into the biology of the broad host range of Pseudomonas aeruginosa phage psØ and indicate that phage has potential for treatment and prevention of infections caused by pathogenic Pseudomonas aeroginosa |
