Abstract

MUC1 is membrane-bound mucin which localizes the apical membrane of normal secretory epithelial cells. It functions in protection and signal transduction of these cells. In cancer cells, MUC1 is abnormally glycosylated and shares in several processes such as angiogensis, invasion and metastasis, immunosuppression and apoptosis.
This study was conducted to immunohistochemically investigate the expression of MUC1 in pleomorphic adenoma and mucoepidermoid carcinoma.
Forty salivary gland neoplasms (twenty benign and twenty malignant) were included in the present study. Using the peroxidase- autoperoxidase method for immunohistochemical examination, each lesion was stained with anti- MUC1 antibody.
Histopathological examination, using hematoxylin and eosin stain, was used to confirm the diagnosis.
Analysis and interpretation of the results was performed using the image analysis software (Image J. 1.41a, NIH, USA) to measure the total mean surface area of immunopositivity for MUC1.
Statistical analysis was performed using ANOVA and Tukey pair wise test for multiple comparaisons of means.
Immunohistochemical results reavealed a higher expression of MUC1 in PA compared to its expression in normal salivary gland tissues. MUC1 is overexpressed in high MEC when compared to low MEC.
Statistical results revealed that there was a statistically significant increase in MUC1 expression in mucoepidermoid carcinoma over pleomorphic adenoma and between high grade mucoepidermoid carcinoma and pleomorphic adenoma, low grade mucoepidermoid carcinoma and pleomorphic adenoma and also between the different grades of mucoepidermoid carcinoma. Also, a significant difference existed between the expression of MUC1 in normal salivary gland tissue and pleomorphic adenoma, between normal salivary gland tissue and low grade mucoepidermoid carcinoma and also between normal salivary gland tissue and high grade mucoepidermoid carcinoma.
from this study it was deducted that MUC1 proved to be a relevant prognostic marker for tumor aggressiveness in benign and malignant salivary gland neoplasms.
