Abstract

Autistic disorder is a pervasive developmental disorder with diagnostic criteria based on abnormal social interactions.
Scientists said that genes play an important role while they have not discovered out which gene or genes are responsible for autism occurrence.
Sometimes in the environment or environmental factors are responsible beside the genetic disturbance for autism occurrence.
Although there is no certain treatment for autism which the good understanding to this disorder leads to the development in the inventional ways to the symptomatic treating methods which relief the severity of the disease so as the child can cope with his life.
The present study was carried out at genetic unit, Faculty of Medicine, Ain Shams University.
The cases and controls were subjected to:
1. History taking sheet that include age, family history of other autistic children, consanguinity of parents, birth difficulties, birth place and medication received.
2. Pedigree construction sheet to find out infections by different disease could be related to autistic infection.
3. Clinical investigation:
- There is no laboratory aid or usage of (X-) ray or any other way to diagnose the disease.
- But the clinical examination depends on the child observation with help of psychiatric and asking the patient parents about his case then to revise already fixed schedules or tables with certain parameters to diagnose the disease.
4. - Evaluation of severity of autistic symptoms using (CARS) test (children autism rating scale).
- It consists of (15) item used to detect the children over (2) years old which are affected with moderatec symptoms.
And this by this scale
(l)  for normal child.
(2)  for mild symptoms.
(3)  for moderate symptoms.
(4)  for severely symptoms.
So children who recorded:
(30 - 33)  Mild.
(34 - 37 )  Moderate.
(37 – and more)  Severe.
Fifth step:
Laboratory investigations
- Measuring level of lead in blood of autistic children.
- Measuring level of Mercury in blood of autistic children.
- (DNA) analysis using (PCR) .
- Glutathione-S-transferase test.
- Pseudocholine esterase test.
Results reveal a noticeable elevation in the level of Lead in blood of number of experimental group patients.
Studied group classified with (CARS) to Mild, Moderate, & Severe (cases) and the results were compared with normal level which is (5 g/dl) which showed no significancy.
The Level of the enzyme of toxicity (pseudocholine esterase) in blood of autistic children under study showed normality by mean results are non significant.
Some showed absence of glutathione-s- transferase enzyme in cases of autistic children under study, its absence means these children have no antioxidant in their bodies, so they will be vulnerable to other infections and many complications.
Dedicated work and continuous act at research field was recommended to discover the genetic causes accused with environmental causes that will guide us to steps which should be taken to reach suitable treatment of this mental and phsyciatric disorder that affects our children.
