Abstract Hepatocellular carcinoma (HCC) is one of the most common cancers worldwide and the most common primary cancer of hepatocytes. More than 80 % of HCC cases occur in developing countries. MGN-3/Biobran is a natural agent obtained by reacting rice bran hemicellulose with multiple carbohydrate hydrolyzing enzymes from Shitake mushrooms. Previous reports have shown MGN-3 to be a potent biological response modifier (BRM) that stimulates several different arms of the immune system including natural killer (NK) cells. MGN-3/Biobran, has been reported to possess strong anticancer effect against several neoplasms. NDEA is one of the most important environmental hepatocarcinogens. It has been reported that a number of animal species developed liver cancer on exposure to NDEA and the posttreatment with CCl4 on the NDEA exerted a strong promoting effect on the development of HCC. The current study was conducted to evaluate the protective role of MGN-3/Biobran against NDEA and CCl4-induced hepatocarcinogenesis in rats and to elucidate the related molecular mechanisms. In this study, liver cancer was induced by the administration of a single dose of NDEA (200 mg/kg BW) i.p. to male albino rats followed by weekly s.c. injections of CCl4 (3 ml/kg b.w./week) for 6 weeks as the promoter of carcinogenic effect. Animals were pretreated or posttreated with MGN-3/ Biobran (25mg/Kg b.w./5 times/week). concluded from the current results that MGN-3/Biobran extracted from rice bran inhibited hepatocarcinogenesis induced by NDEA and CCl4 to a great extent via induction of apoptosis through modulation of the expression of p53, Bax, caspases-3 and Bcl-2 and prevented inflammation and cancer cells proliferation by the inhibition of NF-kB expression and blocking the degradation of IKB-α induced by NDEA-CCl4. |
