Abstract
Imipramine and fluoxetine are from anti depressant drugs .and also used for treatment of other diseases as irritable bowel diseases ,Neuritis ,back pain and nocturnal enuresis in children .
And due to long period of treatment of these drugs which extends to years .it was important to study the effect of them on the human immune system .
The aim of the work is to extract the two drugs (Imipramine and fluoxetine)from serum , urine and hair and to evaluate their effects on immune system .
This study was conducted over 90 persons (60patientsand 30as control group ).patients have been treated in out patient psychiatric clinic in Benisuif university hospital and Al Abbasya psychiatric hospital .from beginning of treatment until 2 years .
The patients are classified according to duration of drug intake into 3 groups :up to 6 months ,from 6 months to 1 year and more than 2years .
In this study , extraction of the drugs was done by different chemical method’s
The extracted drugs was detected by colourtest ,TLC and concentrations measured by HPLC.
Also the effect of the two drugs on the immune system was studied , by measuring total and differential leucocytic count and serum immunoglobulinsIgG and IgM.
The result was as follow :
- First , about colour test which depends on chemical reagents added to the extracted drug and give colours . Marquis and Forrest are specific for Imipramine and gave blue colour ,while did not give any colour with fluoxetine . FPN was not specific either for Imipramine or fluoxetine .
- For TLC , the standard eluent was chlorofrom to methanol ( 4:1) and gave good Rf values . Dragendorff was the best spraying agent .
- About HPLC , it was more accurate and sensitive .
The effect of Imipramine and fluoxetine on the immune system was studied and resulted as follow :
-Decrease in total leucocytic count with Imipramine only with significant P value
-decrease in lymphocytes percentage in differential leucocytic count only with Imipramine ,not with fluoxetine , this decrease become significant with long duration of Imipramine intake.
- there was decrease in serum IgG and IgM with imipramine ,and normal values with fluoxetine .
