Abstract

Cyclophosphamide is commonly used as a cytoreductive agent in the treatment of cancer (blood, breast, and ovary) because of its ability to interfere with DNA synthesis and its pharmacological action on dividing cells. CP also leads to various toxic effects. lipoic acid (LA) is a naturally occurring nutraceutical, whose therapeutic effect has been related to its antioxidant activity and its ability to repair oxidative damage. The main objective of this study is to evaluate the effects of lipoic acid on the toxicity induced by cyclophosphamide in male rats.Rats were randomly divided equally into four groups (I, II, III and IV) of 20 rats per group. The first group (GI) is the control group was received orally distilled water, the second group (GII) was administrated orally with 1.5 mg/kg of cyclophosphamide by oral gavage. The third group (GIII) was received 4.3 mg/kg lipoic acid. The fourth group (GIV) was co-treated with 1.5 mg/kg cyclophosphamide and 4.3mg/kg lipoic acid. Treatments were applied for 28 days. Forty rats (10 per group) were sacrificed after 28 days and the other forty rats were experimented for mating index. No Adverse effects on skin, fur, mucous membranes, and the behavior of animals were observed. A significant decrease in the final body weight was observed after treatment with CP. LA-treated rats did not show any significant variation in body weight. There were no significant differences in the net and relative weights of the liver, kidney, testes and epididymis between the CP, LA and CP-LA groups. CP administration caused significant elevations in total cholesterol, LDL, VLDL, TG and total lipid, and caused significant reduction in HDL levels. A striking elevation of ALT, AST, creatinine, urea, LH and prolactin were observed in CP-treated animals. LA with CP reduced these elevations but it did not return to the control level. LA-treated rats did not show any significant variation. There was a significant decrease in TP, testosterone and FSH levels in CP group. The administration of CP caused a significant decrease in mating and fertility index. CP treatment caused significant decrease in sperm count, motility and viability. LA supplementation alone or together with CP improved the mating and fertility index and caused significant increase in sperm count, motility and viability. The present study has shown that lipoic acid not only ameliorated the CP-induced body weight, AST, ALT, protein, urea and creatinine levels, but also attenuated the changes of the fertility factors, cyclophosphamide has a destructive effect on secretion of sex hormones in male rats and hence effect of lipoic acid improves it.
