Abstract

Asthma is a chronic inflammatory disorder of the airways. It affects an estimated
300 million individuals worldwide and will probably become more than 400 million by 2020 The World Health Organization estimates that13.8 million disability adjusted life years are lost annually due to asthma , representing 1.8% of the total global disease burden. It is estimated that asthma causes 346,000 deaths worldwide every year .The prevalence of asthma is increasing in most industrialized countries especially in children , making it the most common chronic illness of childhood.
The miRNAs are small ∼22 nucleotide noncoding RNAs that inhibit gene expression by mRNA degradation, or translational repression. Importantly, cellular
miRNAs can regulate cell function, including proliferation, differentiation, immune reaction, and tumorigenesis. It is not surprising that circulating miRNAs display consistent profiles between healthy individuals and significantly altered expression in diseases.
miRNAs also have an essential role in both the innate and adaptive immune system and pathogenesis of asthma .
A case - control study was conducted at allergy and pulmonology clinic Beni-Suef
,University Hospital. It was held from April 2016 to September 2016. The study concluded 30 asthmatic children and 20 apparently healthy children .
MicroRNA 221 expression levels were determined by quantitative real-time polymerase chain reaction in all children .
The study concluded the following:
1-Serum Micro RNA 221 levels were higher in all asthmatic patients compared to
controls group with P value ( <0.775 ) .
2-Serum Micro RNA 221 levels were higher in sever asthma(2.25±2.6 ) compared to moderat asthma ,mild asthma intermittent asthma (1.7±1.5 ), (1.4±1.1), (0.91±0.98) respectively.
3-Serum Micro RNA 221 levels were higher in atopic asthmatic patients (1.5±1.5) than in nonatopic asthmatic patients (1.03±0.97) with no significant value .
4-Micro rna levels between asthmatic patients precipitated asthma by smook exposure (1.4±1.4) were higher than in asthmatic patients not precipitated asthma by smook exposure (0.8±0.14) with no significant value .
5-Micro RNA 221 levels were higher in asthmatic patients with positive family history
(1.6±1.5) than asthmatic patients with negative family history (0.8±0.6) with no significant value .
6-Serum Micro RNA 221 levels were higher in all asthmatic patients on corticosteroid therapy with no significant value .
The study concluded that MicroRNA 221 have a role in pathogensis of asthma and in grading of asthma severity and may be of diagnostic and therapeutic value.
