Abstract

Sucralose is an intense artificial sweetener produced by chlorination of sucrose so it is a chlorinated disaccharide, 600 times sweeter than sucrose, and is very stable at high temperatures. It was approved by the FDA, in 1998, to be utilized in foods, beverages, pharmaceutical products and vitamin supplements.
The aim of this study is to assess the effect of sucralose on the liver and kidneys of white male albino rats.
The sample consisted of 50 male albino rats that were provided by Institute of Ophthalmology, Giza. The animals were randomly divided into three groups as follows:
group A: Control group (10 rats): Animals untreated and served as negative control.
group B (20 rats): Sucralose treated group at a dose of 5 mg/kg/day via oral route for 12 weeks.
group C (20 rats): Sucralose treated group at a dose of 125 mg/kg/day via oral route for 12 weeks.
The mean body weight was measured at the beginning and at the end of the study in all different groups. where the highest increase in body weight noted in rats treated by sucralose at high dose recorded 286.00 ± 10.46gm. while the least body weight gain noted in control negative group recorded 247.50 ± 24.86gm.
The variation in liver enzymes (ALT - AST- ALP) and albumin among different study groups are measured where the highest increase of liver enzymes (ALT - AST- ALP) showed with high level in sucralose groups (low dose group &high dose group) recorded (60.65 ± 8.38, 46.80 ± 6.80&246.65 ± 29.54IU/L) in low dose group (71.70 ± 9.57, 46.50 ± 10.67& 193.80 ± 56.86IU/L)in high dose group but insignificant reduction in albumin level in different study groups.
The variation in kidney function tests creatinine and blood urea level among different study groups are measured insignificance difference with in rats’ kidney function tests in different study groups. Urea is 19.2± 1.68 at control group, 19.65± 1.03 at low dose group and 20.25± 1.38at high dose group. Creatinine is 0.69 ± 0.09 at control group, 0.68 ± .093 at low dose group and 0.73 ± 0.14 at high dose group.
The present study represents the hepatic histological changes observed among the different groups where the most affected group which treated by sucralose at high dose showed necrosis and fibrosis.
The renal histological changes observed among the different groups where the most affected group which treated by sucralose at high dose showed focal interstitial inflammatory infiltrate , perivascular inflammatory infiltrate , congestion of the glomerular tufts and focal vacuolation of endothelial lining of glomerular tufts.
