Abstract The present thesis concerned with analytical study of some antihypertensive drugs using chromatographic and electrochemical methods. The studied drugs namely: losartan, irbesartan, valsartan, olmesartan, candesartan, hydrochlorothiazide, amlodipine, nifedipine and atenolol in pure, tablets and spiked human plasma and spiked urine. The thesis comprises an introduction and two main chapters Introduction It is an introductory part which includes general introduction about the studied drugs, their medicinal importance, the chemical structure, and physical properties of the target drugs. The introduction also included a literature review of the official and the reported analytical methods used for the drugs determination. It also included the scope of the research. Chapter I: High performance Thin Layer chromatographic Determination of Some Antihypertensive Drugs Chapter I include three parts, Part1:- selective HPTLC Method for Determination of some Angiotensin II Receptor Antagonists Using Chloranil as Spraying Reagent The first part deals with determination of losartan, irbesartan, valsartan, olmesartan and candesartan by HPTLC after spraying with chloranil for the first time using reflectance/ absorbance mode for quantification. The separation was based on using HPTLC plates pre-coated with silica gel 60 on aluminum sheets as stationary phase. The mobile phase used composed of chloroform : acetone : glacial acetic acid (7.8: 1.5: 0.7 v/v/v) which allowed determination of the studied AIIRAs using single mobile phase. The obtained Rf values for the studied drugs were 0.22 ± 0.01, 0.35 ± 0.015, 0.50 ± 0.005, 0.57 ± 0.01, and 0.70 ± 0.01 for olmesartan, valsartan; irbesartan, losartan and candesartan respectively The responses were linear over the concentration ranges of (400.0 - 1500.0), (400.0. |
