Abstract

This study is designed to evaluate the effect of Acrylamide on the testis of adult albino rat and also evaluate the possible protective effect of Selenium on Acrylamide induced toxicity. Forty adult male albino rats weighting 200-250 gm were used. They were divided into four group, first group served as control, second group (Acrylamide treated group) : received acrylamide in a dose of 50 mg/ kg/ day intra peritoneially for ten days, third group (Selenium treated group) received selenium in a dose of 1 mg/ kg/ day orally for ten days and the fourth group received acrylamide in a dose of 50 mg/ kg/ day intra peritoneially and selenium in a dose of 1 mg/kg /day orally for ten days. After ten days all rats were sacrified and blood samples were taken for assessment of total serum testesterone and caudal epididymis specimens were taken for sperm counting .Also testicular specimens were prepared for paraffin sections and stained with Hx & E and Toluidine blue for light microscopic examination and were prepared for electron microscopic examination. Histopathological and Ultra-structural examination of the testis of acrylamide treated rats showed degenerated and distorted seminiferous tubules with partially or complete absence of sperms. The interstitial tissue showed decrease in number of Leydig cells. Statistical analysis of the weight of rats and weight of testis show significant decrease. Also total serum testosterone level and sperm counting showed significant decrease. However, the testis of acrylamide and selenium treated rats showed less degenerated seminiferous tubules with usual organization of spermatogonia, spermatocytes and sperms inside lumen. There was significant increase in testosterone level and sperm counting of treated rats. Weight of rats and weight of testis also increased. The results of the study were discussed, with a final conclusion of the harmful effect of acrylamide on reproduction that was proved by histopathological and ultra-structure degenerative changes of seminiferous tubules and decreased weight of testis, sperm counting and total serum testesterone and protective effect of selenium on testis was proved by further improvement of sections of testis in acrylamide and selenium treated group.
