Abstract The aim of the present study was to detect brucella in cow’s raw milk shops. Therefore, 200 milk samples were collected from 2 cities (El-Santa city at El-Gharbia Governorate and Sidi-Salem city at Kafer El-Sheikh Governorate) 100 from each throughout one year from February 2016 to February 2017. All samples were examined by conventional assays (Milk Ring Test and Cultural tests) and RT-PCR technique as confirmatory test. The results showed that the positive samples in summer and winter seasons were 12 and 18 by MRT, 0 and 5 by culturing while 13 and 30 by RT-PCR technique respectively. The isolated organisms were identified biochemically and serotyping, the result showed that the isolated organisms were brucella melitensis and this result confirmed by multiplex PCR. All positive 43 RT-PCR samples and isolated strains were examined for detection of virulence genes Ure, bvfA and virB using PCR technique. The result showed that 88%, 79% and 74% of samples for ure gene, bvfA gene and virB gene respectively. Also detection of these virulence genes in DNA extracted from isolates confirmed that brucella species isolated from milk samples were mainly brucella melitensis. |
