Abstract

The current study was out lined to look into the protective effect of the ethanolic extract of moringa oleifera leaves to exhibit hepatoprotective effect against paracetamol induced hepatotoxicity in rats. Thirty six adult albino rats weighting average body weight were allocated into six groups, six rats each .
Biochemical analysis : Intraperitonially injection of paracetamol evoked a significant increase in ALT ,AST ,ALP enzymes ,total bilirubin ,cholesterol with non-significant increase in albumin, globulin, total protein compared with control group, with non-significant decrease in albumin globulin, total protein compared with paracetamol group.
Inflammatory marker: Co administration of moringa oleifera and paracetamol induced a significant decrease in serum C-reactive protein(CRP) concentration.
Antioxidant activities: Administration of moringa oleifera and paracetamol induced a significant increase in catalase , super oxide dismutase , glutathione peroxidase concentration compared with paracetamol group.
Oxadative stress marker: Co administration of moringa oleifera and paracetamol induced a significant decrease in malondialdehyde (MDA), nitric oxide (NO) concentration.
Comet assay: The present study was undertaken to test the in vivo anti genotoxic effect of moringa oleifera in rats using the single cell gel electrophoresis (comet assay), administration of Paracetamol alone caused significant increase in comet tail length and % of DNA.
Histopathology: Examination of liver tissues obtained from rats intraperitonially injected with paracetamol alone revealed hydropic degeneration and fatty change in large number of hepatocytes. The portal area showed mild congestion of hepatic arterioles and venules , dilated lymphatic, mild biliary proliferation and moderate round cells infiltration. Multifocal necrosis of hepatocytes and partial replacement of the hepatocytes by erythrocytes were seen. .
Apoptic Markers: Liver tissues obtained from rats intraperitonially injected with paracetamol alone revealed mild apoptotic reactions with 3-5% of cells were affected for liver .The anti apoptotic reaction showed very weak positive reaction 1-2% of hepatocytes .
