Abstract

The purpose of this study was to evaluate the effect of local injectionof autologous PRP on orthodontic tooth movement in rabbits. This studywas conducted on 25 white male New Zealand rabbits categorized into fivegroups according to the time of sacrifice. The rabbits were sacrificed at the1st, 7th, 14th, 21st and 30th day. Split mouth control was used and the right sidewas randomly selected as the experimental side while the left side was set ascontrol. Autologous PRP of 0.4 ml was prepared and injected equally in thesubmucosa in the facial, lingual, mesial and distal to the lower right firstmolar. One hundred gram of orthodontic force by NiTi closed coil springbetween the lower first molars and lower incisors in both sides of the lowerjaw. The distance between the distal contact point of the first molar and themesial contact point of the second molar on both sides of the lower jaw wasmeasuredusing precise digital orthodontic gauge. Tissue sections was fixedin 10% buffered formalin, decalcified in EDTA solution then embedded inparaffin sections and processed forexamination by routine Hematoxylin andEosin stain and examined under light microscope. All the collected datasubjected to statical analysis.In the present study the OTM was statically insignificant difference inthe study side in comparison with the control side in the first 7 days of theexperiment. However, the OTM in the experimental side was significantlyhigher than the control side in the 14th, 21st and the 30th day. Histologically,in the control side there was hyaline areas appeared in the periodontalligament at the first 7 days which was not seen in the study side. In the otherhand there was an abundant blood vessels in the experimental side at fromthe first day and continue to appear in the all experimental periods.In thestudy side the osteoblastic and osteoclastic activities were higher than thecontrol sideat the 14th, 21st and the 30th day. The conclusion was that the local injection of PRP could accelerate the OTM in rabbits.
