Abstract

The compounds investigated throughout this work were determined in laboratory prepared mixtures. These compounds include ethinyl estradiol (EED), testosterone (TST), 17α-methyltestosterone (17MTST) and progesterone (PRG).
The thesis consists of two main chapters
Chapter 1
This chapter is divided into three main parts
A- General introduction to the pharmacological effects of the studied compounds, their structures, physical properties as melting point, molecular formula, molecular weight and solubility in different solvents.
B- General introduction to environmental and food analysis which includes some related definitions, role of analytical chemistry in environmental and food analysis, main challenges in environmental and food analysis, different sources of hormones into the environment, effects of the long term low dose exposure of environmental hormonal residues
C- General introduction to sample preparation and extraction methods which include solid phase extraction, it’s types, steps, advantages disadvantages and liquid liquid extraction.
Chapter 2
This Chapter is divided into two main parts
A- General introduction to sources of hormones in the selected matrices which are hen egg, chicken liver and Tilapia pond fresh water sample.

B- A fast isocratic HPLC-UV method was presented for the simultaneous determination of EED, TST, 17MTST and PRG. HPLC separation and quantitation were made on 150 x 4.6 mm (i.d) Agilant C18 reversed phase. The mobile phase used was 70% methanol: 30% 0.1 % formic acid by volume. The flow rate was maintained at 1.00 mL\ min. the mobile phase was filtered through 0.45-μm disposable filter. All determinations were performed at 25 ºC. the injection volume was 20 μL. Quantitation was achieved using UV detection at 220 nm based on peak area. The method showed good precision, high accuracy, high extraction recovery and wide linear range.
Confirmation of the applicability of the developed method was performed on real hen egg samples, chicken liver and Tilapia pond fresh water sample. Liquid chromatography tandam mass spectrometry was used for the determination of hormonal residues of EED, TST, 17MTST and PRG in real samples; hen egg, chicken liver and Tilapia pond fresh water sample.
