Abstract

This study was done to make typing of S. aureus by clumping factor genes (clfA and clfB) which act as cell surface proteins that play important roles in the pathogenesis of S. aureus caused disease. They thought to be essential for colonization and establishment of infections. Also, antimicrobial resistance of S. aureus became on the top of the serious problems that have a negative impact on public health. Recently, the wide spread of antibiotic resistance stimulates studies to search for new methods on treatment with plant extracts and essential oils.
Therefore, the main objective of this study is to make typing of S. aureus by clumping factor genes and to extend the research to evaluate the antibacterial activities of N. sativa oil on S. aureus in addition to assessing the impact of their subinhibitory concentrations on the transcriptional modulation of S. aureus cell wall genes (mprF and graS).
Out of 100 samples collected from mastitic cow milk from different farms in Sharkia and Ismalia Governorates, 63(63%) staphylococcal isolates (30 (30%) S. aureus and 33 (33%) CoNs) were recovered.
Antibiotic susceptibility testing of S. aureus isolates revealed that majority of isolates were MDR with the highest sensitivity rate against Imepenim(100%) and marked resistance patterns against β-lactam antibiotics. It is worth noting that.
