Abstract

The present work investigates some pharmacodynamic and the possible adverse effects of doramectin andoxyclozanide given each alone and concomitantly administered to mature male rats . Experiments were carried out to investigate the effects of these drugs on blood picture, some biochemical parameters , semen characters and serum testosterone levels in mature male rats. Moreover, the effects of these drugs on the histological structure of the liver, kidney and the reproductive organs of treated and control mature male rats were investigated .
For this experiment 60 mature male rats were divided equally into 4 groups each of 15 rats as follows:
group I:Rats were injected with doramectin (0.3 mg/kg b.w. s.c).
group II: Rats were given oxyclozanide(20mg/kg b.w orally).
group III:Rats were injected with doramectin (0.3mg/kg b.w. s.c) + given oxyclozanide (20 mg/kg b.w orally).
group IV: Rats were given 2ml saline /kg b.w orally and kept as a control group. Dose of each drug was calculated according to the method described by Paget and Barnes (1964).All drugs were readministered to all groups after 30 days from the start of the experiment as the same doses previously mentioned. Five rats from each treated and control groups were killed after 2nd ,4th and 8th week from the beginning of drug administration. Blood, body organs and epididymal contents were obtained from treated and control rats.
The obtained results are as follows :
HaematologicalFindings :
The obtained results showed that theadministration of doramectin andoxyclozanide given each alone and concomitantly administered to mature male ratsproduced improvement inHb % in doramectin treated group at 4th week.However administration of the anthelmentics produced a significant decrease in RBCs, W.B.Cs. counts and hematocrite value
Biochemical Findings :
The obtained results showed that the administration of doramectin and oxyclozanide given each alone and concomitantly administered to mature male rats produced a significant increase in serum of AST, ALT and ALP . There was an increase in serum bilirubin levelin male rats treated with doramectin alone at 8th week and there was significant increase of serum albumin level at 4thweek with doramectin alone and doramectin togother oxyclozanide.
Hormonal Findings :
The administration of doramectin and oxyclozanide given each alone and concomitantly administered to mature male rats produced a significant decrease in serum testosterone level .
Reproductive Findings :
The obtained results showed that the administration of doramectin and oxyclozanide given each alone and concomitantly administered to mature male rats produced significant effects in sperm concentration, motility, abnormalities) and the testes weight and epidedymis weight .
The administration of doramectin and oxyclozanide given each alone and concomitantly administered to mature male rats produced a significant decrease in the weight index of the reproductive organs,theliver and the kidney .
Histo-pathological Findings :
