Abstract

We hypothesize that acetyl salicylic acid acts as antidiabetic drug through activation of PPAR-γ and inhibition of NF-κβ and pro-inflammatory gene expression so, this work aim to investigatethe biochemical and molecular mechanisms related to the antidiabetic effect of both low and high doses of acetyl salicylic acid through assessment the effect of acetyl salicylic acid on PPAR-γ and NF-κβ . Also, investigate the beneficial effects from combination of acetyl salicylic acid with another antidiabetic drug especially PPAR-γ agonists.To achieve that the following protocol was followed :
Seventy male adult albino rats weighing 150-200g were subdivided into 2 groups. The first one served as a normal control group (G1) (10 rats), the rats were supplied with the typical rat chow in addition to pure water. The other 60 rats were fed high fat and high fructose (HFHF) diet for 12 weeks.after 12 weeks, rats with blood glucose level less than 200mg/dl were excluded and considered as non-diabetic. Only rats which had blood glucose levels >200 mg/dl were considered resistant diabetic after confirmation of insulin resistance (IR) using a homeostasis model assessment of insulin resistance ( HOMA-IR).
