Abstract

This study was carried out to investigate the modulating effect of vitamin E (100mg/kg b.wt.) and/ or ginseng (200mg/kg b.wt.) on adverse effect of levofloxacin (10mg/kg b.wt.) for 14 successive days.
This experiment was done on fifty apparently healthy adult male albino rats. The first blood sample was collected with anticoagulant as EDTA for hematological studies to determine erythrogram and leukogram. The second blood sample was collected without anticoagulant to obtain clear serum for biochemical analysis.
Effects on haematological parameters: Oral administration of vitamin E and/ or ginseng 2 hours before levofloxacin for 14 successive days in male rats displayed a significant increase in total erythrocytic count, Hb content, PCV % and Plt count with a significant decrease in TLC and DLC.
Effects on serum biochemical parameters:A) Liver enzymes: Administration of vitamin E or ginseng 2 hours before levofloxacin displayed a significant decrease in ALT, AST and ALP activities.
B) Proteins profile: Administration of vitamin E or ginseng 2 hours before levofloxacin elicited a significant increase in total proteins and albumin levels.
C) Lipids profile: Oral administration of levofloxacin in male rats elicited a significant increase in total cholesterol, TG, LDL and VLDL levels with a significant decrease in HDL levels while oral administration of vitamin E or ginseng 2 hours before levofloxacin evoked a significant decrease in total cholesterol, TG, LDL and VLDL with a significant increase in HDL levels.
D) Kidney functions: Administration of vitamin E or ginseng 2 hours before levofloxacin in male rats displayed a significant decrease in urea and creatinine levels.
E) Antioxidants/Oxidant status: Oral administration of levofloxacin in male rats produced a significant increase in MDA concentration with a significant decrease in CAT and SOD activities while oral administration of vitamin E or ginseng 2 hours before levofloxacin displayed a significant decrease in MDA concentration with a significant increase in CAT and SOD activities.
F) Tumor necrosis factor-α Non-significant variation in TNF-α were recorded post administration of ginseng in levofloxacin treated male rats.
The Histopathological investigation: Oral administration of vitamin E 2 hours before levofloxacin revealed an enhancing effect of vitamin E in liver sections. Kidney sections showed enhancement in the renal lesions where most of the nephrons units were normal.
