Abstract

This study was carried-out for-
1- Isolation and identification of Pseudomonas spp. from human (pus & urine) and from animals mastitis milk.
2- In vitro antimicrobial sensitivity test for Pseudomonas.
3- Ability of the isolated strain to form biofilm.
4- Detection of some virulence factors (ESBL) using ERIC-PCR.
This study was carried-out on different samples that includes
A. MASTITIS Milk samples:-
Milk samples collected from 4 cows clinically suffering from mastitis.
B. Human samples that includes:
5 Pus samples of wounds and 6 urine samples of human with UTI that collected from humans at the private hospitals that suffering from chronic wounds have pus.
The samples transported to laboratory and in the laboratory were undergoing cultivated on the different cultural media, microscopical examination, and antibiotic sensitivity tests. Identification of ESBL- genes examinations phenotypically and genoyypically was done by ERIC PCR and similarity testing.
We found that:-
1- Pseudomonas aerogenosa showed characteristic bluish-green pigmentation on cetrimid agar, grew on MacConkey’s agar media, without lactose fermentation and on nutrient agar it produces circular mucoid smooth colonies which emits sweat grape or tortilla odour . .
2-On the Microscopic characters indicated that, they were Gram-negative pink rods.
3-The higher incidences of Pseudomonas aerogenosa observed in Pus samples, followed by urine samples and the least observed in the mastitis milk samples.
3-While, results on the Antibiotic sensitivity test cleared that, in PUS samples the higher sensitivity observed for the antibiotics of COL Meanwhile, the results on the Urine samples cleared that, the antibiotic sensitivity test for the bacteria isolated from pus showed a higher sensitivity observed for the antibiotics of COL (100 %) while, the sensitivity for SCF and CIP reached to 66.67% and the sensitivity of AK, TOB, IMP, PTZ, MEM and CAZ) showed a moderate sensitivity level as its level reached to (50 %).
4- The ESBL-gens detection cleared that, the the CX-CX and CIAT test gave+ve results for ampC ESBL Pseudomonas aeruginosa strains. The mCIM test and IPM EDTA test gave +ve results for Metalo β lactemase ESBL Pseudomonas aeruginosa strains.
5-Meanwhile, the results on the PCR cleared that, the Electrophoresis Image of PCR Genotyping of Pseudomonas aeurogenosa Lane L, 16 DNA molecular size marker (100 - 3000bp) ) Show that each cluster isolates showing similar ERIC profile, which indicated clonal similarities between the isolates .
6-While, the results on the similarity index (Jaccard / Tanimoto Coefficient and number of intersecting elements) between all samples cleared that, the distribution of the genes and distances between the studied genes in all samples have a good homogenity and similarity between the samples.
