The impact of Micro-RNA level on therapeutic outcome in colorectal cancer patients

Abstract
Introduction: Colorectal cancer (CRC) is one of the leading causes of mortality and morbidity in the world. The commonly used methods for CRC diagnosis are not enough sensitive, specific, and invasive, therefore, there is a steady need for the development of CRC-specific, noninvasive markers to be used for diagnosis, prognosis, and response to treatment. Therefore, this work aimed to explore the role of miRNAs (223-3p, 20a-5p, 19a-3p, 17-5p, and 7-5p) as non-invasive screening biomarkers for CRC detection, prognosis, and prediction of response to the treatment as well as to explore their roles on target genes EGFR, ERK and PTEN. Methods: In this study 77 naïve CRC patients and 20 normal healthy individuals were used for comparing their miRNAs expression levels using realtime PCR to test their sensitivity and specificity for the diagnosis of CRC. After 3 and 6 months of FP-based therapy, blood samples were withdrawn from each patient to determine the expression levels of miRNAs after treatment. In addition, the protein levels of studied miRNA- targeted genes, EGFR, ERK, and PTEN, were determined at baseline, as well as after 3 and 6 months of treatment by using ELISA technique. All patients were followed up for 5 years to calculate their 5 years OS and clinical outcome
