Abstract

Gold nanoparticles (GNPs) have several biomedical applications such as photothermal therapy, drug delivery, targeting, and diagnosis of cancers. GNPs have great compatibility with antibodies, antigens, and other biomolecules. Therefore, surface functionalization of GNPs can be used to accelerate the antibody-antigen reaction. In this study, anti-EGFR (Anti-epidermal growth factor receptor) was conjugated to GNPs then labeled with FITC (Fluorescein Isothiocyanate). The Anti-EGFR-GNPs-FITC was characterized by UV-VIS, Zeta potential, and TEM. The Anti-EGFR-GNPs-FITC was applied for targeting tumor-induced in hamster Mucosal Cells using Confocal Laser Scanning Microscope (CLSM). CLSM study reveals the successful distribution and GNPs bioconjugation
and accumulation in tumor cells. In addition, a real-time PCR study showed an increase in mean values of P53 (tumor suppressor gene) expression in both normal and tumor-induced cells. The up-regulation in gene expression of P53 means using Anti-EGFR-GNPs-FITC will help in suppressing the tumor cells growth. This study reveals that Anti-EGFR-GNPs-FITC was successful in molecular and confocal laser scanning microscopic evaluation of mucosal cells.
